The route is decided by the molecule

How a compound is made is part of how a supplier is judged. The choice is between solid phase, solution phase and a hybrid of the two.

Three routes

  • Solid phase. The chain is built on a resin. It loses yield as the chain grows, because closely related impurities get harder to remove.
  • Solution phase, also called liquid phase. More scalable and cheaper at volume, and not well suited to larger peptides.
  • Hybrid. Short segments are made on resin and joined in solution.

Where the choice matters most

Cyclic peptides and difficult small molecules are where the choice matters most. All three approaches are used; the right one depends on the molecule.

Makers and resellers

A supplier who can explain which route they would choose, and which building block they would order, makes the compound. One who cannot is reselling.
