What goes wrong at the supplier

Most of this market’s material comes from contract laboratories. The patterns below are what we have seen in our own supplier work. They are observations, not a statement about every laboratory, and they are the reason a certificate alone is not enough.

Five patterns

What happensHow it shows upHow we catch it
Purification is cut short: fewer passes, or the salt exchange skippedClosely related impurities sit under the main peak and the purity figure looks better than the material is. Counter-ion levels stay high.A second, slower HPLC method, the impurity profile by LC-MS, and a counter-ion measurement
Synthesis steps go wrong: an incomplete coupling, the wrong building block, a ring that did not closeMissing-residue sequences, a different compound altogether, or the open form of a cyclic peptideLC-MS against the correct expected mass, and NMR for structure
Isomers form during synthesisD-isomers that behave almost identically in a standard testChiral purity testing
Data is read wrongPurity taken from a method that cannot separate the impurities. Mass “confirmed” against the wrong expected value. Content assumed to equal purity. We read the raw chromatograms and spectra ourselves, not the summary line
A reseller presents itself as the makerNobody can answer questions about route, building blocks or impuritiesAsk which route and which building block. Makers answer; resellers deflect.

Why a certificate is not enough

Verifying this from outside is hard. There is no access to the plant, the certificate is issued by the party being checked, and one good batch says little about the next. That is why every batch is tested independently and every supplier is scored over time, not once.
